Protective Effect of Epigallocatechin-3-Gallate in Hydrogen Peroxide-Induced Oxidative Damage in Chicken Lymphocytes

Figure 4

Intracellular ROS and SOD. Lymphocytes () were treated with EGCG (0, 22.5, 45, and 90 μM) for 6 h first and then cultured in media with (model) or without (normal control) H2O2 (80 μM) for 4 h, then dyed for ROS and SOD. Cells with increased production of SOD showed orange fluorescence and were detected with the FL2 channel. Cells with increased production of ROS demonstrated green light and were registered in the FL1 channel. (a) Flow cytometric gating percentage; (b) H2O2 + EGCG 90 μM group; (c) H2O2 + EGCG 45 μM group; (d) H2O2 + EGCG 22.5 μM group; (e) H2O2 (model) group; (f) normal control group; (g) ROS levels represented by mean green fluorescence intensity in the indicated groups; (h) SOD levels represented by mean orange fluorescence intensity in the indicated groups. Data are presented as (). Bars with different letters are significantly different ( or ).
