Sirt5 Attenuates Cisplatin-Induced Acute Kidney Injury through Regulation of Nrf2/HO-1 and Bcl-2

Sirt5 regulates mitochondrial morphology and ROS production. (a) Mitochondrial status (fusion/fission) was examined by Mitotracker staining. Cisplatin treatment induced mitochondrial fragmentation/fission. Sirt5 overexpression reduced mitochondrial fragmentation while Sirt5 depletion increased mitochondrial fragmentation. (b) Cellular ROS production was examined by CellRox staining. Cisplatin treatment elevated ROS level. Sirt5 overexpression reduced ROS production while Sirt5 depletion increased ROS level in HK-2 cells treated with cisplatin. The fluorescence intensity indicating ROS level was shown in the bar chart. .
