Sirt5 Attenuates Cisplatin-Induced Acute Kidney Injury through Regulation of Nrf2/HO-1 and Bcl-2

Figure 1

Sirt5 HK-2 metabolic activity treated with cisplatin. (a) Expression of Sirt5, Nrf2, HO-1, and Bcl-2 proteins in HK-2 cells treated with 0, 5, and 10 μg/mL cisplatin for 24 hours. (b) Protein and mRNA expression of Sirt5 in HK-2 cells transfected with Sirt5 plasmid and siRNA. (c, d) MTT assay showed that Sirt5 overexpression increased metabolic activity while Sirt5 siRNA decreased metabolic activity when treated with different concentrations of cisplatin (0, 2.5, 5, and 10 μg/mL) at different time points (24 and 48 hours). (e) Sirt5 overexpression increased HK2 growth rate. Sirt5 depletion decreased cell growth rate. .
