Effects of Aminoglycoside Antibiotics on Human Embryonic Stem Cell Viability during Differentiation In Vitro

Figure 3

Treatment with antibiotics affects the expression of neural stem cell markers. (a) Cells (passage number 43) were collected for RNA isolation on day 10 of differentiation in the presence of antibiotics as indicated and qRT-PCR was performed to analyze the expression of Pax6, Emx2, Otx2, and Pou3f2 using gene-specific primers. The expression of Polr2g gene was used to normalize the expression. The bars represent normalized fold mRNA expression with values of untreated samples set as 1. The expression of Polr2g gene was normalized to the expression of beta-actin gene. Error bars represent standard deviation. , , and . (b) The protein levels of Pax6 was analyzed using Western blot for samples treated with antibiotic concentrations as indicated. The expression of GAPDH was used as loading control. The results are representative of at least three independent differentiation experiments.
