Age-Dependent Accumulation of 8-Oxoguanine in the DNA and RNA in Various Rat Tissues

LC-MS/MS chromatograms of dGsn and 8-oxo-dGsn derived from DNA. DNA samples obtained from tissues were denatured and hydrolyzed to nucleosides by successive treatments with nuclease P1 and alkaline phosphatase. To each sample, [15N5]dGsn and 8-oxo-[15N5]dGsn were added to provide appropriate internal standards. The mixtures were applied to the LC-MS/MS system for quantification of the two types of deoxyguanosine, dGsn and 8-oxo-dGsn. The upper two figures show the elution patterns of DNA hydrolysates and the lower figures represent those for the internal standards.
