JNK and NADPH Oxidase Involved in Fluoride-Induced Oxidative Stress in BV-2 Microglia Cells

JNK/MAPK signal pathway is involved in fluoride-induced oxidative stress of BV-2 cells. (a) BV-2 cells were treated with 1, 5, 10, and 50 mg/L NaF for 24 h, and cell lysates were separated on 10–20% SDS-PAGE and analyzed by Western blot. (b) and (c) Intracellular DHE and DAF-FM fluorescence intensity for and NO contents were measured by flow cytometer in BV-2 cells preincubated with 50 μM JNK/MAPK inhibitor (SP600125) for 2 h, followed by 50 mg/L NaF for 24 h. (d) BV-2 cells were pretreated with 2 mM MEL for 2 h, followed by 50 mg/L NaF for 24 h, and cell lysates were blotted with antiphospho (p)-JNK or anti-JNK antibodies.
