Expression of iNOS mRNA over time and effects of SB202190 and BIRB 796 on the expression of iNOS mRNA in response to cytokine mixture in A549 cells. (a) Cells were stimulated with cytokine mixture (CM: TNF, IFNγ, and IL-1β; 10 ng/mL each) for the time indicated, and total RNA was extracted. The expression of iNOS mRNA was determined by quantitative real-time RT-PCR, and mRNA expression was normalized against GAPDH mRNA. Unstimulated cells (0 h) were set as 1, and other values were related to that. Results are expressed as mean ± S.E.M., . One-way ANOVA with Dunnett’s posttest was performed, and statistical significance is indicated with as compared to unstimulated cells. (b) Cells were preincubated with SB202190 (1 μM) or BIRB 796 (100 nM) for 30 min, stimulated with cytokine mixture for 6 h, and harvested for total RNA extraction. The expression of iNOS mRNA was determined by quantitative real-time RT-PCR, and mRNA expression was normalized against GAPDH mRNA. Results are expressed as a percentage of CM, mean ± S.E.M., . One-way ANOVA with Bonferroni’s posttest was performed, and statistical significance is indicated with as compared to cells treated with CM.
