Antibody-Hapten Recognition at the Surface of Functionalized Liposomes Studied by SPR: Steric Hindrance of Pegylated Phospholipids in Stealth Liposomes Prepared for Targeted Radionuclide Delivery

Analysis of MAb 734 binding on tethered DSPC/DSPE-DTPA-indium bilayer created on CM5 chip and of MAb 734 binding to DTPA-indium-tagged liposomes adsorbed onto a L1 chip via (1/Ms), (1/s), Rmax (RU), and (M) parameters. MAb 734 was prepared in two-fold serial concentration series (720 nM to 2.185 nM) and was injected at a flow rate 60 μL/min across the liposomes adsorbed L1 chip surface for 3 min. At the end of each injection, the dissociation phase was set to 4 min. Liposomes binding assay was only performed once with CM5 and duplicated with L1 chip. and values were obtained by nonlinear regression of experimental data fitted with the SCK mathematical model. The score of χ 2 (Chi2) is <5; it means that the model used adequately describes our data. , , and values are the average ± standard deviations.
