Improving the Catalytic Activity of Hyperthermophilic Pyrococcus horikoshii Prolidase for Detoxification of Organophosphorus Nerve Agents over a Broad Range of Temperatures

Prolidase assays were performed in triplicate at 10°C, 20°C, 35°C, 50°C, 70°C, and 100°C and contained 0.2 mM CoCl2 and 4 mM Met-Pro. WT-Ph1prol-specific activity was 66 U/mg at 10°C, 131 U/mg at 20°C, 246 U/mg at 35°C, 340 U/mg at 50°C, 973 U/mg at 70°C, and 3,955 U/mg at 100°C.
