Nuclear-Targeting Delivery of CRISPRa System for Upregulation of β-Defensin against Virus Infection by Dexamethasone and Phenylalanine Dual-Modified Dendrimer

In vitro and in vivo relative β-defensin mRNA expression quantified by real-time PCR analysis in the human lung adenocarcinoma A549 cells (a), human kidney 293T (b), mouse neuroblastoma (in vitro), and BLAB/C mice (in vivo) (c). Dose: 1 μg/μL dCas9 plasmid and 1 μg/μL sgRNA plasmid were complexed with G4 (Phe)50%-DET/plasmid at N/P of 8 and Lipo2000 at N/P of 2. Inhalation dose: 5 μg/μL dCas9 plasmid and 5 μg/μL scr sgRNA or 5 μg/μL sgMBD4 in 100 μL PBS. Mouse neuroblastoma N2a cells were used in in vitro experiments. BLAB/C mouse administrated with complexes was tested. MBD4 and HBD1 are abbreviations of mouse beta-defensin-4 and human beta-defensin-1, respectively. The scr sgRNA represented nonsense sgRNA and sgMBD4 represented mouse β-defensin-4 sgRNA (sg2+sg3) with the highest expression at mRNA level. Data are (). , .
